NTHi肺炎小鼠肺2型与3型脱碘酶mRNA的变化

         

摘要

Objective:To explore the changes of T3 , T4 and TSH levels in serum and type 2 iodothyronine deiodinase(D2) and type 3 iodothyronine deiodinase(D3) mRNA levels in lungs of mice of nontypeable haernophilus influenza(NTHi) pneumonia, to speculate on mechanisms of pneumonia and euthyroid sick syndromes(ESS) and to provide theoretical basis to the treatment. Methods:C57BL/6 mice were randomly divided into control group, 3 days after infection group, 7 days after infection group and 10 days after infection group. Blood samples and lungs were taken 3,7 and 10 days after establishment of NTHi pneumonia model, T3 , T4 and TSH levels in serum were detected by radioimmunoassay method and D2 and D3 mRNA levels in lungs were measured by real time polymerase chain reaction(Real-time PCR). The data were statistically analyzed by SPSS13. 0 software package. Results: There were statistical differences of T3 ,TSH,D2 and D3 mRNA levels among the 4 groups (F=47. 129,9. 028,16. 600,10. 505, while there was no statistical difference of T4 levels among the 4 groups (F=0. 467). T3 and TSH levels of 3 days after infection group were lower than control group, and then picked up gradually, there was no statistical differences between 10 days after infection group and control group; correspondingly, D3 level of 3 days after infection group was higher than control group and D2 level of 7 days after infection group was higher than control group and then gradually dropped, but both were still higher than control group 10 days after infection, the differences were statistically significant. Conclusion; ESS may occur as complication of NTHi pneumonia and changes of D2 and D3 mRNA profiles involve in it.%目的:探讨不可分型流感嗜血杆菌(NTHi)肺炎小鼠血三碘甲状腺原氨酸(T3)、甲状腺素(T4)、促甲状腺素(TSH)及肺2型脱碘酶(D2)与3型脱碘酶(D3)mRNA水平的变化,推测肺炎并正常甲状腺病态综合征(ESS)的发生机制,为其治疗提供理论依据.方法:C57BL/6小鼠随机分为对照组、感染3d组、感染7d组与感染10d组.对照组取血及肺组织,感染组于制备小鼠NTHi肺炎模型后3、7及10d取血及肺组织,放射免疫分析法检测血清T3、T4及TSH水平,实时定量聚合酶链反应(Real-time PCR)法测定肺D2及D3 mRNA水平.数据以SPSS13.0统计软件进行统计分析.结果:四组T3、TSH、D2及D3 mRNA相对表达水平的差异有统计学意义,T4的差异无统计学意义.感染后3d T3与TSH水平低于对照组,后逐渐回升,于感染后10d其水平与对照组差异无统计学意义;与之相对应,感染后3d D3水平高于对照组,感染后7d D2水平方始高于对照组,后逐渐回落,于感染后10d仍均高于对照组,差异有统计学意义.结论:NTHi肺炎可并发ESS,D2与D3 mRNA水平的变化与其发生有关.
