一株野生侧耳属菌株的分离鉴定与生物学特性

         

摘要

采用组织分离的方法从野生侧耳属菌株(Pleurotu ssp.)子实体上分离得到菌株 XZG -ts,以其基因组为模板扩增获得该菌株的 ITS 序列,序列分析结果表明,菌株 XZG -ts 与 GenBank 中来自于澳大利亚、印度和中国云南的3个肺形侧耳菌株聚为一簇,组内核苷酸一致率为100%,而与侧耳属其他4个种的10个菌株核苷酸一致率为98.10%~99.06%,依此结果将 XZG -ts 菌株鉴定为 P.pulmonarius。进一步研究了不同碳源、氮源、碳氮组合、温度及 pH 值对菌株 XZG -ts 菌丝生长的影响,结果表明,最适碳源为果糖,氮源为酵母膏,最佳碳氮源组合为果糖1%、麦芽糖1.0%、酵母膏0.2%、蛋白胨0.3%,最适生长温度为25℃,最适 pH值为6。%A fungi strain,XZG -ts,collected from the Mountain Tai,was isolated by tissue isolation and the internal transcribed spacers (ITS)was amplified by PCR.In the phylogenetic tree constructed with ITS,XZG -ts formed a branch with 3 isolates belonging to Pleurotus pulmonarius from Australian,India and China with the consistent rate of 100% in intra -group nucleotide.While it was only 98.10% ~99.06% with other 10 isolates of Pleurotus.Therefore XZG -ts was identified as P.pulmonarius.The effects of carbon source,nitrogen source,carbon -nitrogen combination and pH value on the growth of mycelium were further studied.The results showed the optimum carbon source,nitrogen source,temperature and pH value was fruc-tose,yeast extract,25℃ and 6,respectively.And the optimum carbon -nitrogen combination was 1% fruc-tose,1.0% maltose,0.2% yeast extract,0.3% peptone.
