Objective To investigate the effect of nerve growth factor (NGF) and its receptor tyrosine protein kinase A (TrkA) in brain astrocytoma. Methods A total of 48 cerebral astrocytoma specimens (glioma group) and 48 tumor adjacent normal brain tissues (normal group) were involved in this study. Immunohistochemical method was used to detect NGF and TrkA expression of two groups. RT-PCR was used to detect NGF and TrkA mRNA relative expression. Relationship between NGF and TrkA protein, mRNA, clinical pathology parameters was assessed. Speraman rank correlation analysis was used to analyze correlation of NGF, TrkA protein and mRNA expression. Results The positive rate of NGF protein in glioma group and normal group were 77% , 31% , respectively (P<0.05). The positive rate of TrkA protein in glioma group and normal group were 85% , 43% , respectively(P<0.05). NGF mRNA relative expression level in glioma group and normal group were 0.78 ± 0.19, 0.41 ± 0. 12, respectively (P < 0.05). TrkA mRNA relative expression level in glioma group and normal group were 0.83 ±0.04, 0.51 ±0.13, respectively (P<0.05). The protein and mRNA expression of NGF and TrkA were correlated with pathological grade but gender and age. NGF, TrkA protein were positive correlated with mRNA expression. Conclusion NGF and TrkA may play roles in the occurrence and development of astrocytic tumor.%目的 探讨神经生长因子(NGF)及其受体酪氨酸蛋白激酶A(TrkA)在人脑星形胶质细胞瘤发生、发展中的作用.方法 选择脑星形胶质细胞瘤组织标本48份(胶质瘤组),瘤旁正常脑组织48份(正常组),采用免疫组化法检测两组NGF和TrkA蛋白表达,RT-PCR法检测NGF和TrkA mRNA相对表达量,并分析NGF和TrkA蛋白、mRNA表达与脑星形胶质细胞瘤临床病理参数的关系,采用Speraman等级相关分析法分析NGF、TrkA蛋白和mRNA表达的相关性.结果 胶质瘤组及正常组NGF蛋白表达阳性率分别为77％、31％,TrkA蛋白表达阳性率分别为85％、43％,两组比较P均＜0.05；胶质瘤组及正常组NGF mRNA的相对表达量分别为0.78±0.19、0.41±0.12,TrkA mRNA的相对表达量分别为0.83±0.04、0.51±0.13,两组比较P均＜0.05.NGF和TrkA蛋白、mRNA表达与脑星形胶质瘤病理分级相关,而与性别、年龄无关.NGF、TrkA蛋白和mRNA表达量呈正相关.结论 NGF和TrkA在星形胶质细胞瘤的发生、发展过程中可能起促进作用.
