脐带华通胶间充质干细胞对急性再生障碍性贫血T淋巴细胞亚群的影响

         

摘要

目的 体外观察脐带华通胶间充质干细胞(UCWJMSCs)对急性再生障碍性贫血(AAA)患者T淋巴细胞(TLC)亚群的调节作用.方法 收集21例AAA患者和10例健康志愿者外周血,制备单个核细胞(MNCs)悬液,UCWJMSCs和MNCs以一定比例混合共育一定时间后,采用流式细胞仪检测TLC亚群并进行统计分析.结果 AAA患者TLC亚群呈Th1、Tc1极化状态.混合培养后,健康人的TLC亚群未见明显变化(P>0.05);AAA患者的TLC亚群除Th2与Tc2无明显变化外,CD+3、CD+8、CD+28、Th1和Tc1亚群比例明显降低,而CD+4、CD+4/CD+8、CD+56、CD+25和CD+103亚群比例明显升高(P<0.05).调节作用24 h就已产生,72 h时达高峰;调节作用随UCWJMSCs浓度的增高而增强.结论 UCWJMSCs对AAA患者的TLC亚群有明显调节作用,不同个体来源的UCWJMSCs调节作用无区别.%Objective To investigate the modulatory effects of umbilical cord Wharton' s jelly mesenchymal stem cells (UCWJMSCs) on T lymphocyte (TLC) subsets in cases with acute aplastic anemia (AAA) in vitro. Methods Peripheral blood samples collected from 21 cases with AAA and 10 healthy volunteers were processed for mononuclear cells (MNCs) suspension. UCWJMSCs and MNCs were mixed at a certain ratio and cocultured for 0-120 h. TLC subsets were detected by FCM post-coculture and analyzed statistically. Results AAA was a disease with Th1 and Tc1 polarized status. There were no changes in TLC subsets in healthy volunteers post-coculture. But TLC subsets in AAA cases changed markedly except for Th2 and Tc2, i.e. the percentages of CD3+, CD8+, CD28+, Th1 and Tc1 subsets decreased markedly, and that of CD4+,CD4+/CD8+, CD56+, CD25+ and CD103+ subsets rised significantly (P <0. 05). The modulatory effects produced 24 h and peaked 72 h post-coculture. The modulation depended upon the UCWJMSCs concentration. Conclusion UCWJMSCs could significantly modulate TLC subsets in AAA cases, but UCWJMSCs derived from different individuals played the same role in the modulation.
