青蒿琥酯对口腔扁平苔藓T-bet和GATA-3平衡影响的研究

         

摘要

目的 探讨青蒿琥酯(artesunate,ART)对口腔扁平苔藓(oral lichen planus,OLP)患者外周血转录因子T- be/GATA-3失衡表达的调节作用.方法 通过密度梯度离心法分离OLP患者外周血单个核细胞(peripheral blood mononuclear cells,PBMC),分别用不同浓度(0、1、100 μmol/L)的青蒿琥酯和5%的碳酸氢钠溶液作用于PBMC 24 h,逆转录聚合酶链反应(RT -PCR)测定法和免疫印迹法(Western blotting)检测OLP患者PBMC中T- bet和GATA -3 mRNA和蛋白的表达.结果 T- betmRNA和蛋白的表达在100μmol/L青蒿琥酯组(0.54±0.26、0.93±0.40)均低于对照组(1.35±0.63、1.47±0.43)(P值均<0.01),GATA-3 mRNA和蛋白的表达在1μmol/L(0.62±0.33、1.11 ±0.69)和100 μmol/L(0.23±0.21、0.44±0.27)青蒿琥酯组中均低于对照组(1.40±0.66、1.68±0.61)(P值均<0.01).结论 青蒿琥酯能够调节OLP患者T- bet和GATA -3的失衡表达.%Objective To investigate the effects of artesunate on expression of the T cell-specific transcription factors T-bet/GATA-3 in oral lichen planus ( OLP) . Methods Peripheral blood mononuclear cells ( PBMC) were isolated by density gradient centrifugation from OLP patients. The cells were treated with different concentrations of artesunate(0,1,100 μmol/L) and 5% sodium bicarbonate for 24 hours. Expression of T-bet and GATA-3 mRNA and protein in the PBMCs was detected by reverse transcription-polymerase chain reaction ( RT-PCR) and western blotting respectively. Results The expression of T-bet mRNA and protein in l00μmol/L artesunate (0.54 ±0.26,0.93 ±0.40)was weaker than that in the control group(1.35 ±0. 63,1. 47 ±0. 43) (P <0. 01 ,P <0. 01). The expression of GATA-3 mRNA and protein in lμ.mol/L(0.62 ±0.33,1. 11 ±0.69) and 100 μm01/L.(0.23 ±0.21,0.44 ±0.27)artesunate was weaker significantly lhan that in the control group(1.40 ±0. 66,1. 68 ±0. 61 ) ( P <0. 01 ,P < 0. 01 ,P <0. 01 ,P <0. 01 ). Conclusions Artesunate can rectificate the imbalance of transcription fautors T-bet and GATA-3. Arlesunate may be an option in the treatment of OLP.
