土茯苓组方对急性肝衰竭大鼠保护作用的实验研究

         

摘要

Objective To observer the effects of smilax prescription on expressions of Bcl-2,Caspase-3 and cytokines in rats with acute liver failure and to verify its exact effect and the possible mechanism. Methods Forty Wistar rats were divided into normal group,model group,smilax prescription group and acetylcysteine group. The smilax prescription group was given smilax prescription solution 10mg/(kg·d),the acetylcysteine group was given acetylcysteine 50mg/(kg·d),and 3 days consecutively. 1 hour after the last time,the acute liver failure model was reproduced in rats by peritoneal injection of D-Galactosamine(D-GalN)and lipopolysaccharide(LPS). After 6 hours,the liver tissue of rats were taken out for Western Blot assay of Bcl-2 and Caspase-3 protein expressions,and TNF-α,IL-6,IL-10 in serum were detected by immunohistochemistry. Result Compared with the normal group,Bcl-2 protein expressions were significantly decreased in the liver tissue of model group,and Caspase-3 protein expressions were significantly increased,the TNF-α、IL-6、IL-10 in serum were same to(P<0.05). Compared with the model group,Caspase-3 protein expressions and TNF-α,IL-6,IL-10 in the group of smilax prescription and acetylcysteine were significantly down-regulated,while Bcl-2 protein expressions were significantly up-regulated(P<0.05). Compared with the acetylcysteine group,Caspase-3 protein expressions and TNF-α,IL-6,IL-10 in the group of smilax prescription were slightly up-regulated,Bcl-2 protein expressions were slightly down-regulated(P>0.05).Conclusion Smilax prescription has protective effect on the apoptosis of rats with acute liver failure,and its possible mechanism may be related to up-regulated of Bcl-2 protein expressions,down-regulated of Caspase-3 protein expressions to and intervention proinflammatory cytokines and anti-inflammatory cytokines.%目的 观察土茯苓组方对急性肝衰竭(ALF)大鼠Bcl-2和Caspase-3蛋白及细胞炎症因子表达的影响,验证其确切疗效及防治急性肝衰竭的作用机制.方法 40只Wistar雄性大鼠随机分为正常组、模型组、土茯苓组及乙酰半胱氨酸组.土茯苓和乙酰半胱氨酸组造模前3d分别予土茯苓水煎液5ml/(kg·d)及乙酰半胱氨酸50mg/(kg·d)灌胃(土茯苓水煎液含生药浓度2g/ml,乙酰半胱氨酸溶液浓度为1mg/ml),采用D-氨基半乳糖(D-GalN)/内毒素脂多糖(LPS)腹腔注射造成大鼠ALF模型.造模6h后,取血检测血清天冬氨酸转氨酶(AST)、丙氨酸转氨酶(ALT)、总胆红素(TBIL)含量,采用酶联免疫法(ELISA)检测TNF-α、IL-6、IL-10,并开腹取大鼠肝组织,应用Western Blot实验方法分别检测肝组织中Bcl-2和Caspase-3蛋白表达情况.结果 与正常组比较,模型组大鼠血清AST、ALT、TBIL、TNF-α、IL-6、IL-10含量和Caspase-3蛋白表达均明显升高(P<0.05),Bcl-2蛋白表达明显下降(P<0.05);与模型组比较,土茯苓组和乙酰半胱氨酸组血清AST、ALT、TBIL、TNF-α、IL-6、IL-10含量和Caspase-3蛋白表达显著下降(P<0.05),Bcl-2蛋白表达明显显著升高(P<0.05);与乙酰半胱氨酸比较,土茯苓组血清AST、ALT、TBIL、TNF-α、IL-6、IL-10含量和Caspase-3蛋白表达略高(P>0.05),Bcl-2蛋白表达略低(P>0.05).结论 土茯苓可明显减轻急性肝衰竭大鼠肝脏损伤,其作用机制可能与上调Bcl-2蛋白、下调Caspase-3蛋白,从而抑制细胞凋亡,以及干预促炎因子及抗炎因子有关.
