丝素蛋白取向电凝胶仿生诱导羟基磷灰石复合物合成的实验研究

         

摘要

Objective In the present study,we used the silk fibroin gel in orientation as template to induce the synthesis of hydroxyapatite in SBF. We have investigated the influence of different concentrations and pre-mineralization temperature on the surface appearance and calcium/phosphorous ratio. Methods Three kinds of different concentrations silk fibroin gel in orientation were prepared by electrophoresis which were put into pre-mineralization water for three times and into SBF for 24h. We had investigated the influence of different kinds of preminilization temperature on the surface appearance. Results The surface appearance transform three-dimensional hole lamellar structure from small-sphere compared with silk fibroin gellike hydroxyapatite. The structure and surface appearance of the apatite were investigated by FTIR,XRD,SEM and ICP. The Results show that the HA crystal grows along the c-axes with increase of the concentration and preminilization temperature. And the HA morphology in the form of self-assembled on the gel surface transferred to three-dimensional hole lamellar structure. Conclusion The silk fibroin gel in orientation was used as template in the study, then calcium and phosphonium ions nucleation and growth on surface of silk fibroin gel in orientation,The composite material are similor to bones and teeth on composition,structure and morphology. So,the silk fibroin gel in orientati-on/HA will take important part in providing a high performance repair material for tissue engineering,plastic surgery and dental fields.%目的:用丝素蛋白取向电凝胶作为模板在模拟体液中仿生诱导羟基磷灰石的合成,并系统研究不同浓度的取向电凝胶和预钙化温度对所合成材料的表面形貌和钙磷摩尔比的影响。方法制取三种不同浓度具有一定取向性的丝素蛋白电凝胶,经过预钙化3次后分别放入模拟体液中静置24h,并研究不同预钙化温度下复合材料表面的形貌特征变化。结果通过改变取向电凝胶浓度和改变预钙化温度,与未经过电场作用的类凝胶仿生矿化诱导生成的羟基磷灰石比较,表面形貌发生了明显的变化。并对所得材料进行测试表征,复合材料的钙磷摩尔比与人体骨组织和牙体硬组织非常接近。结论以丝素蛋白取向电凝胶作为模板,钙磷离子在精确的调控和引导下,所制备的复合材料在组成、结构上及形态上与人体骨组织和牙体硬组织相似。有望为组织工程、整形外科和牙科领域提供一种性能优良的修复材料。
