外周血中性粒细胞凋亡相关蛋白质对SAP的诱导凋亡机制研究

         

摘要

ObjectiveTo investigate the mechanism of apoptosis induced by peripheral blood neutrophile granulocyte apoptosis related proteins and somatostatin in patients with severe acute pancreatitis. Method 90 SD rats were randomly divided into the severe acute pancreatitis group (SAP)and the sham operation group(SO),45 rats in each group,to detect the peripheral blood neutrophile granulocyte survivin expression, neutrophile granulocyte survivin protein expression and expression of neutrophil survivin mRNA by theMethodsof TUNEL fluorescence labeling method,immunohistochemical method and the RT-PCR method.ResultsSAP Group compared with SO group,24h,48h,72h three point neutrophil apoptosis rate decreased obviously(P<0.05),the SO group at different time points of neutrophil apoptosis rate had no significant difference (P>0.05);The peripheral blood neutrophils espressed survivin in two group,and the SAP group was higher than that in the SO group at the time of 24h,48h and 72h(P<0.05),the peripheral blood neutrophils expressed survivin in SO group,there is no significant difference(P>0.05),The peripheral blood neutrophils expressed survivin in two group,and the So group was higher than that in the SAP group at the time of 24h,48h and 72h (P<0.05),the peripheral blood neutrophils expressed survivin in SAP group,there is no significant difference(P>0.05).ConclusionThere is a close relationship between neutrophil apoptosis and SAP development,to curb the further development,improve the prognosis of the patients. Survivin is plays a regulatory role in the delay of neutrophil apoptosis.%目的:探讨外周血中性粒细胞凋亡相关蛋白质及生长抑素在重症急性胰腺炎(SAP)时的诱导凋亡机制。方法将90只SD大鼠编号,随机分为重症急性胰腺炎组(SAP组)和假手术组(SO组),每组45只,使用TUNEL荧光标记法、免疫组织化学法、RT-PCR法分别检测大鼠的外周血中性粒细胞凋亡情况、中性粒细胞survivin蛋白表达情况和中性粒细胞survivin mRNA的表达情况,分析比较两组的检测结果。结果 SAP组与SO组比较,24h、48h、72h三个时点中性粒细胞凋亡率呈现明显的下降(P<0.05),SO组各时点中性粒细胞凋亡率差异无统计学意义(P>0.05);SO组和SAP组大鼠外周血中性粒细胞survivin蛋白均有表达,SAP组大鼠24h、48h、72h三个时点表达明显高于SO组各相应时点(P<0.05),SO组各时点中性粒细胞survivin 蛋白均有表达,差异无统计学意义(P>0.05);SO组和SAP组中性粒细胞survivin mRNA有表达,但SAP组24h、48h、72h组中性粒细胞survivin mRNA表达明显高于SO组(P<0.05),SO组各时点中性粒细胞survivin蛋白均有表达,差异无统计学意义(P>0.05)。结论中性粒细胞的凋亡与SAP发展有密切关系,能遏制其进一步发展,改善患者预后。Survivin与中性粒细胞凋亡延迟有关,对其起到调控作用。
