目的：观察穿心莲内酯干预对哮喘大鼠气道IL-33表达的影响，探讨其作用机制。方法 SD大鼠36只随机分成3组：哮喘组、穿心莲内酯干预组和对照组，每组各12只。通过100mg卵清蛋白（OVA）腹腔注射致敏，并且OVA雾化吸入连续激发28d，末次激发24h后处死大鼠。行支气管灌洗液（BALF）细胞总数、嗜酸性粒细胞计数，AB-PAS染色观察气道组织杯状细胞、黏液分泌情况，Real-time PCR检测IL-33 mRNA在肺组织内的表达。结果穿心莲内酯干预组BALF细胞总数、嗜酸性粒细胞数、AB-PAS阳性染色面积、IL-33 mRNA表达均较哮喘组减低（P<0.05）。结论穿心莲内酯可抑制哮喘大鼠气道炎症，并且降低气道IL-33的表达。%Object To evaluate the effect of andrographolide on expression of IL-33 in a rat model of asthma.MethodsSD rats(n=36)were randomly divided into three groups:the normal group(n=12),the asthmatic group(n=12),the andrographolide intervention group(n=12). The rats were sensitized by 100mg ovalbumin(OVA),then they were challenged with ovalbumin,24 hours after the fi nal inhalation and challenging,all the rats were sacrifi ced. The amount of total cells and the concentration of eosinophils in the bronchoalveolar lavage fl uid(BALF)of each group were determined,the metaplasia of goblet cells in airway wall along with the degree of mucus hypersecretion were observed by alcian blue/periodic acid schiff (AB-PAS)staining. The pulmonary expression of IL-33 mRNA were measured by real-time PCR.ResultsThe amount of total cells,eosinophils in BALF,the airway infl ammatory pathological score,the goblet cells in the airway wall,the degree of mucus secretion,the level of IL-33 mRNA in andrographolide group were all signifi cantly lower than those in asthmatic group(P<0.05).ConclusionThe andrographolide can suppress the airway infl ammation and decrease the expressions of IL-33.
