血小板源性生长因子C在家兔视网膜静脉阻塞模型的表达研究

         

摘要

目的:研究家兔视网膜静脉阻塞(RVO)模型血小板源性生长因子C(PDGF-C)表达、作用及参与RVO新生血管形成的相关机制。方法通过氩激光光凝建立视网膜静脉阻塞家兔模型,观察结束后分别在1周、2周及4周三个时相点处死家兔,采取标本,观察病理改变,免疫组化法观察PDGF-C蛋白表达情况。结果与空白对照组比较,RVO家兔模型视网膜组织中各时相(1周、2周和4周组)PDGF-C蛋白表达均有不同程度的增高,差异有统计学意义[(4.833±1.835)pg/mL、(10.500±1.634)pg/mL、(5.167±1.835)pg/mL比(2.167±0.408)pg/mL,P<0.01],其中2周模型组与1周、4周模型组比较差异有统计学意义(P<0.01),1周模型组与4周模型组比较,P=0.713。结论PDGF-C在家兔视网膜静脉阻塞血管内皮细胞中呈现高表达,预示其可能在RVO发生发展中起重要作用,且其表达程度与视网膜缺血具有一定的时间相关性。%Objective To observe the expression of platelet-derived growth factor-C (PDGF-C) in the retina of rabbits with retinal vein occlusion(RVO) , and to investigate the effect of PDGF- C in RVO and the possible mechanism in the pathogenesis of retinal neovascularization. Methods The RVO of rabbit model was established with argon laser photocoagulation. After the 1st week, 2ed week, and 4th week, the rabbits were sacrificed to ob-serve the pathological changes by H-E staining and the expression of PDGF-C protein by immunohistochemistry. Results Compared with control group, the expression of PDGF-C protein in RVO rabbit model was significantly higher after the 1st, 2 nd, and 4th week, with a statistically significant difference(4.833 1.835 pg/mL, 10.500 1.634 pg/mL, 5.167 1.835 pg/mL vs 2.167 0.408 pg/mL, P<0.01). A significant difference in the expression of PDGF-C protein was found between 2nd week and 1st and 4th week(P<0.01), but there was no significant differ-ence between 1st week and 4th week(P=0.713). Conclusion PDGF-C is highly expressed in vascular endothelial cells of rabbits with RVO, indicating that it may play an important role in the development of RVO, and that its expression level has a certain time correlation with retinal ischemia.
