参麦注射液对大鼠肢体缺血再灌注损伤保护作用的实验研究

         

摘要

目的:探讨参麦注射液对大鼠肢体缺血再灌注损伤的保护机制。方法30只SD大鼠随机分为3组,每组10只:缺血再灌注对照组(对照组)、缺血再灌注参麦组(参麦组)采用扎闭股动、静脉缺血4h,开放再灌注1h制造缺血再灌注模型,并分别于再灌注即刻给予0.9%氯化钠注射液、参麦注射液4 ml/kg腹腔注射;假手术组不结扎股动静脉,4h后给予0.9%氯化钠注射液4 ml/kg腹腔注射。腹主动脉取血,予以抗凝离心取上清液,测定总超氧化物歧化酶(T- SOD)活性及丙二醛(MDA)、一氧化氮(NO)、内皮素-1(ET-1)含量。结果与假手术组比较,对照组T- SOD活性降低,MDA含量升高(P<0.01)。参麦组T- SOD活性较对照组升高, MDA含量降低,NO及ET-1含量降低(均P<0.01),而NO/ET-1增大(P<0.01)。结论参麦注射液可减少骨骼肌缺血再灌注损伤后大鼠机体自由基生成,提高自由基清除酶活性,改善再灌注损伤血管内皮功能紊乱,对缺血再灌注损伤有一定的防治作用。%Objective To investigate the protective effect of Shenmai injection on skeletal muscle fol owing limb ischemia reperfusion injury in rats. Methods Thirty SD rats were randomly divided into three groups (n=10 in each group): Ischemic reperfusion+NS group (I/R group), Ischemic reperfusion+Shenmai injection group (SM group) and sham- surgery group(S group). Ischemic reperfusion were induced by ligation of rat femoral vessels. The plasma T- SOD, MDA, NO and ET- 1 were de-termined. Results As compared with S group, the T- SOD activity in I/R group was significantly lower (P<0.01) and MDA con-tent was significantly greater (P<0.01). Compared with I/R group the T- SOD activity in SM group was significantly higher (P<0.01) and MDA content was significantly lower (P<0.01). Compared with I/R group,plasma levels of ET- 1 and NO in SM group were decreased (P<0.01) and the NO/ET- 1 ratio was increased (P<0.01). Conclusion Shenmai injection can resist lipid per-oxidation of free radicals and protect the endothelial cells, which may be clinical y used for the ischemia- reperfusion injury.
