There is considerable interest in understandingnhow multiple binding events can be mediated through ansingle protein interface. Here, a synthetic library approachnwas developed to generate a novel dual-specific antibody.nUsing a combinatorial histidine-scanning phage display library,npotential metal binding sites were introduced throughoutnan anti-RNase A antibody interface. Stepwise selectionnof RNase A and metal binding produced a dual-specificnantibody that retained near wild-type affinity for its targetnantigen while acquiring a competitive metal binding site thatnis capable of controlling the antibodyu0001antigen interaction.nStructure analysis of the original antibodyu0001RNase A complexnsuggested peripheral interface residues and loop flexibilitynare key contributors for obtaining the dual specificity.
