TheDNA interstrand cross-link (ICL) resultingnfrom the C40n-oxidized abasic site (C4-AP) is a uniquenclustered lesion comprised of a cross-link adjacent to annick. The ICL is a substrate for the UvrABC nucleotidenexcision repair system. The strand containing the nick isnpreferentially incised, but the nick influences the cleavagensites. Moreover, in approximately 15% of the molecules,nthe strand opposite the nick is incised, resulting in a morentoxic double-strand break. This is the first example innwhich an interstrand cross-link is converted by nucleotidenexcision misrepair into a more deleterious double-strandnbreak.
