In Sepia officinalis, egg-laying consists of successive stereotyped steps in the following sequence: (1) ovulation, (2) embedding, (3) fertilization, (4) attachment of the egg to a suitable substrate. Study of the cuttlefish in the natural environment, in captivity and in the lab has clarified some of the egg-laying mechanisms. New peptides are identified by means of purification methods, in vitro biotests and mass spectrometry analysis. Among these peptides, the SepOvotropin or PKDSMLLLQVPVYamide was characterised, which modulated contractions of the whole genital tract from a threshold concentration of 10~(-19) M as opposed to 10~(-14) M for ILME. The latter released by the oocyte in the female genital tract and by the egg in the environment, is the first pheromone characterised in a cephalopod mollusc. Another peptide involved during an ultimate steps of egg-laying was also isolated: the PIDPGVamide or SepSAP. This oocyte factor, swich attracts spermatozoa at concentrations near 10~(-17)M, is the first sperm-attracting peptide identified in protostomians. The ensemble of these peptidic factors provides a preliminary framework for regulatory system of egg-laying, emphasising the complexity of this physiological mechanism.
