Recently, more attention has been attracted by intelligent drug delivery systems (DDS), because the release of drug can be controlled via the system feedback responding to pathologic environment signals, Using natural polymers such as chitosan to compose this kind of pulsed DDS will be of benefit, because chitosan [α(1→4)2-amino-2-deoxy-β-D-glucan] is one of the abundantly available polysaccharides in nature. As we know, chitosan is usually obtained by alkaline deacetylation of chitin, the principal component of shells of living organisms, e.g. crab and shrimp. Chitosan has low toxicity, is biocompatible and can reduce lipid level. Nishimura et al. have loaded adriamycin to chitosan beads and studied their antitumour activity Chandy et al. have used chitosan as a matrix for oral sustained delivery of ampicillin. The present authors have developed a pH-responsive semi-interpenetrating polymer network hydrogel of chitosan and polyether. The pulsed-release behavior of chlorhexidini acetas and cimetidine is studied. Considering the antiacid and antiulcer characteristics of chitosan, in this work, a hybrid polymer network (HPN) microspheres of chitosan and gelatin were synthesized via co-crosslinking them with glutaraldehyde, and an investigation was made on the pH-sensitive pulsed-release behavior of cimetidine-loaded microspheres for weakening the side reaction of the drug for the treatment of duodenal and benign gastric ulceration.
