Theobjectiveofthisstudywastodeterminewhetherasimple,noninvasivemethodinvolvingadministrationofisoproterenolcouldbeusedtoproducemyocardialinjuryandcardiacdysfunctioninthemouseheartwithalowincidenceofmortality.AdultSwiss#8211;Webstermicewereinjectedwithisoproterenol(100mg/kgSC)oncedailyfor5d.Myocardialhistologyandleftventricular(LV)functionwereassessed10to14dafterthelastisoproterenolinjectionin14survivingisoproterenol-treatedmiceand15saline-treatedcontrolmice.Leftventricularsystolicanddiastolicpressureswereevaluatedinvitrobymeansofisovolumicallycontracting,perfusedLangendorffpreparations.Isoproterenolinducedmarkedendocardialinjury,associatedwithhypertrophyofsurvivingmyocytes,andanincreaseinmyocardialfibrosis(collagentypesIandIIIaccordingtopicrosiriusredmicroscopy).Theheartsfromisoproterenol-treatedmicedemonstrateddecreasedLVcompliance,asevidencedbyanupwardshiftinthediastolicpressure#8211;volumerelationship,withnormalLVsystolicfunction.Isoproterenoladministrationprovidesasimple,noninvasivemeanstoinduceendocardialinjuryanddiastolicdysfunctionwithoutsignificantimpairmentofsystolicfunction.Thismodelhasalowincidenceofmortalityandmaybeusefultoassesstheeffectsofgeneorstemcelltherapyoncardiacdysfunctionwithoutthepotentialconfoundingeffectsofinvasiveprocedures.
