Severalstudieshaveindicatedastrongassociationbetweenasthmaandaspirationofstomachcontents.However,thecomplexassociationbetweentheseinflammatoryprocesseshasnotbeenstudiedextensivelyinanimalmodels.Inthepresentstudy,wedevelopedananimalmodeltoevaluatetheinflammatorycell,chemokine,andairwayresponsestoasthmacomplicatedbyaspiration.Themodelwasproducedbysensitizingmicetocockroachallergensfromhouse-dustextracts.Micewithasthma-likeairwayresponsesthenwereinoculatedintratracheallywitheitheranacidicsolutionorsaline.Acidaspirationincreasedairwayhyperresponsivenessinmicewithasthmaforatleast8h.After6h,thecombinedinjurycausedanadditive,notsynergistic,increaseinairwayhyperresponsivenessandneutrophilrecruitmenttotheairways.Althoughcysteinylleukotrienesinbronchoalveolarlavagefluidwerehigherafteracidaspiration,treatmentwithareceptorantagonistbeforeaspirationdidnotdiminishairwayhyperresponsiveness.Vagalmechanismsreportedlymediateairwayresponsesinacidaspiration;however,pretreatmentwithananticholinergicagentdidnotreduceairwayresponsestoacid.Theseresultsareconsistentwithaneffectivemodeloftheacuteeffectsofaspirationontheallergiclung.Furtherstudiescouldexaminehowvariousformsofaspirationinfluencetheseverityofasthma.
