4-Vinylcyclohexenediepoxide(VCD)causesearly,gradualovarianfailureinmicebecauseitspecificallytargetssmallpre-antralovarianfollicles.Theperiodbetweenlossofthesefolliclesandovarianfailureisanalogoustoperimenopauseinwomen.WesoughttocharacterizetheperiodofonsetofovarianfailureinVCD-treatedmiceinregardtoestrouscyclelengthandhormonalchanges.FemaleC57Bl/6mice(age,28days)weredoseddailyfor15dayswithVCD(160mg/kgintraperitoneally)tocauseearlyovarianfailureorwithvehicleonly(controlanimals).Cyclelengthwasmonitoredbyvaginalcytology.Plasmalevelsof17#59137;-estradiol(E2),progesterone(P4),andfollicle-stimulatinghormone(FSH)incontrolandVCD-treatedanimalsweremeasuredduringproestrusofcycles1through12.Cyclelength(mean,5.8days)didnotdifferbetweengroupsforcycles1through4.Incontrast,cyclelengthduringcycles5through12wasincreased(meanlength,10.9days;P0.05versuscontrol)inVCD-treatedanimals,whichalsoshowedanapparentincreaseinplasmaFSHlevels.PlasmaE2andP4atproestrusdidnotdifferbetweengroupsduringanycycle.OvarianfailureinVCD-treatedmicewasconfirmedbyhistologicalevaluationonday156afteronsetofdosing,whereascontrolanimalswerestillcycling.Therefore,despitecompromisedcyclelengthinVCD-treatedmice,peakovariansteroidproductioninpreovulatoryfolliclesatproestrusisadequate.TheseresultsdemonstratethattheVCD-treatedmousecanserveasanappropriatemodeltomimichormonalchangesduringtheperimenopausaltransitioninwomen.
