Wepresentanewperfusionsystemandsurgicaltechniqueforsimultaneousperfusionof2tissue-isolatedhumancancerxenograftsinnuderatsbyusingdonorbloodthatpreservesacontinuousflow.Adult,athymicnuderats(Hsd:RH-Foxn1suprnu/sup)wereimplantedwithHeLahumancervicalorHT29colonadenocarcinomasandgrownastissue-isolatedxenografts.Whentumorsreachedanestimatedweightof5to6g,ratswerepreparedforperfusionwithdonorbloodandarteriovenousmeasurements.Thesurgicalprocedurerequiredapproximately20mintocompleteforeachtumor,andtumorswereperfusedforaperiodof150min.Resultsshowedthattumorvenousbloodflow,glucoseuptake,lacticacidrelease,Osub2/subuptakeandCOsub2/subproduction,uptakeoftotalfattyacidandlinoleicacidandconversiontothemitogen13-HODE,cAMPlevels,andactivationofseveralmarkerkinaseswereallwellwithinthenormalphysiologic,metabolic,andsignalingparameterscharacteristicofindividuallyperfusedxenografts.Thisnewperfusionsystemandtechniquereducedproceduretimebymorethan50%.Thesefindingsdemonstratethat2humantumorscanbeperfusedsimultaneouslyinsituorexvivobyusingeitherrodentorhumanbloodandsuggestthatthesystemmayalsobeadaptedforuseinthedualperfusionofotherorgans.Advantagesofthisdualperfusiontechniqueincludedecreasedanesthesiatime,decreasedsurgicalmanipulation,andincreasedefficiency,therebypotentiallyreducingthenumbersoflaboratoryanimalsrequiredforscientificinvestigations.
