Circadianclocksorganizeawidearrayofmetabolicfunctionsinacoherentdailyscheduleandensuresynchronyofthisschedulewithenvironmentalrhythms.Dailyrhythmicityoflipidmetabolismoccursinrodentsandruminants.Weexamineddailylevelvariationsofserumlipids(nonesterifiedfattyacids[NEFA],triglycerides,phospholipids,totalcholesterolandtotallipids)inhealthydogs,particularlyfocusingontheirtemporalrelationshiptolightingandfastingcycles.WhereasserumNEFAlevelsdidnotchangeacrosstheday,levelsoftotallipids,totalcholesterol,phospholipids,andtriglyceridesoccurredindogsmaintainedunder12:12-hlight:darkcyclesandfedasinglemealdaily.Onlytherhythmicpatternoftriglyceridesrespondedtoa6hdelayinlightonset,suggestingacardinalroleofalight-entrainedcircadianoscillatorinitsgeneration.Toinvestigatewhethertemporalvariationsinserumlipidsdependtophysiologicalpostprandialchanges,wemeasuredlipidlevelsinfasteddogs.Rhythmsoftotallipids,totalcholesterol,phospholipids,andtriglyceridesvanishedwhendogswerefood-deprived,indicatingthattheserhythmsaredrivenbythedigestiveprocess.LevelsofserumNEFApatternsweresignificantlyhigherduringfastingthanafterfoodintake.TheincreaseofNEFAconcentrationsduringfastingmayreflectthemobilizationofadiposetissueNEFAmediatedbythedecreaseininsulinwithitslypoliticeffects.Elucidatingthedailyrhythmicityoflipidlevelsisfundamentaltounderstandingthemetabolismofthedog,ananimalmodelfrequentlyusedforresearchinmetabolicpathophysiology.
