Oraltoleranceisdependentonthecomplexarchitectureofthemucosalsystemofthegastrointestinaltract,itsassociatedlymphoidtissue,andspecializedimmunecells.Changesinthisarchitectureorthefailureofanyofitscomponentsmayhinderthegenerationoforaltolerance.Thelarynxandesophagusarethegatewaytothegastrointestinaltract,servingasthesiteoforalantigenintroductiontotheimmunesystemandmayhaveanimportantroleinestablishingtolerance.Intragastricgavageisacommonmethodforpreciseoraldosingofrodents,particularlyinstudiesexaminingoraltolerance.However,complicationssuchasesophagealtraumacanoccurandinducecomplicatingfactorsthataffectexperimentaloutcomes.Inthisstudy,weexaminedtheesophagealepitheliumforalterationsresultingfromlong-termrepeateddailyuseofintragrastricgavageanditseffectontheinductionoftolerance.Tolerancetoovalbumincouldnotbeachievedafterusingintragastricgavagefor14dormoreconsecutivelytointroduceovalbumin.However,tolerancewasachievedwhenintragastricgavagewasusedforshorterdurations.After14dofgavage,disruptionoftheesophagealmucosalepitheliumindicativeofaninflammatorypathology,cellularinfluxintotheesophagealtissue,andproinflam-matorycytokinesinthetissuewereabsent,andtheCD3sup+/supcellpopulationintheesophagealepitheliumdecreased.Thesefindingsprovideinitialevidencefortheimportantrolesofesophagealintegrityandcellularpopulationsintheinductionoforaltoleranceandsuggestpossibleimmunologicsequelaeinexperimentsinvolvingtheuseofextended,repeatedgavage.
