BackgroundandPurpose:EnterotoxigenicEscherichiacoliheat-stableenterotoxin(STa)isamajorcauseofdiarrheainyounganimals.Age-dependentvariationinthedensityandaffinityofthemouseenterocytereceptorsspecificforSTawasinvestigated.brxmlns="http://pub2web.metastore.ingenta.coms/"/brMethods:Fouragegroups(2-day-,1-and2-week-,and2-month-old)ofSwissWebstermicewerestudied(8to10mice/group).FlowcytometryandradiolabeledSTa(125I-STa)assayswereusedasreliablequantitativemeasuresforcharacterizationofSTa-enterocytereceptorinteraction.brxmlns="http://pub2web.metastore.ingenta.coms/"/brResultsandConclusions:InteractionofSTawithitsputativereceptorwasstrongerforenterocytesof2day-oldmice.Scatchardanalysisof125I-STa-receptorinteractionsuggestedthatSTa-receptorsexistathighernumbersonenterocytesfrom2-day-old(7.2nmol/mg)thanolder(0.30,0.36,and0.40nmol/mgfor1-week-,2week-,and2-month-oldmice,respectively).Additionally,receptorsfrom2-day-oldmicehadgreateraffinityforSTa(Ksubxmlns="http://pub2web.metastore.ingenta.coms/"d/sub=75nM)thandidreceptorsfromoldermice(Ksubxmlns="http://pub2web.metastore.ingenta.coms/"d/sub=125,1,430,and1,111nMfor1-week-,2-week-,and2-month-oldmice,respectively).DensityofSTareceptorsonenterocytesandtheiraffinitytoSTamaydetermineextentofbindingandseverityofthesecretoryresponse,andmayexplainthehighsusceptibilityofnewbornanimalsandhumaninfantstoSTa-mediateddiarrhea.
