Repeateddailydosingofmicewith4-vinylcyclohexenediepoxide(VCD)causesagradualonsetofovarianfailure,providingamodelforperimenopause.Becauseincreasingnumbersofwomenaredelayingstartingafamily,infertilityinagingwomenisofconcern.ThisstudywasdesignedtodeterminetheeffectsofimpendingovarianfailureonfertilityinVCD-treatedmice.FemaleC57BL/6Jmiceweredoseddaily(17d)withvehiclecontrolorVCD(160mg/kg,intraperitoneally)todepleteprimordialfolliclesandthenweredividedinto2groups.Group1wasmatedsoonafterdosing;group2wasmatedonday20afterdosing,duringimpendingovarianfailure.Fertilitywasevaluatedongestationalday16.Ingroup1,cyclelength,pregnancyrate,andnumberoflivefetusesdidnotdifferbetweenVCD-treatedanimalsandcontrols,butVCD-treatedmicerequiredmorematingstobecomepregnantandhadmoreresorptions.Ingroup2,VCD-treatedmicedemonstratedproestrusandcopulatoryplugs,butonly1animalbecamepregnant,andshehadnoviablefetuses.Ovariesfrompregnantandnonpregnantcontrolscontainedsimilarnumbersoffolliclesandcorporalutea.OvariesfromVCD-treatedanimalscontainednofollicles,andcorporaluteawereseenonlyinpregnantanimals.InVCD-treatedmicematedsoonafterdosing,conceptionwasmoredifficultandmoreresorbedfetuseswereseen,whereasinthosematedclosertoimpendingovarianfailure,nosuccessfulpregnancieswereachieved.TheseresultsdemonstratethatVCD-treatedmicecanbeusedtomodelinfertilityinperimenopausalwomen.
