Insulinpromotesearlyembryonicdevelopment,butwhetherthisactionaffectspostimplantationfetaldevelopmentandalterstheexpressionofimprintedgenesremaintobedetermined.ThisstudyanalyzedtheexpressionandmethylationlevelsofthegrowthrelatedimprintedgenesH19andinsulin-likegrowthfactor2(Igf2)infetusesexposedtoinsulinbeforeimplantation.Wecultured2-cellembryosineither0or0.25#59157;g/mlinsulinuntiltheblastocyststageandthentransferredthemintopseudopregnantrecipientmice.Thenumberofembryosdevelopingtoblastocystsafterinsulinexposurewas16.4%higherthanthatofthecontrol,andthebirthbodyweightoftheinsulin-exposedgroupwas17.8%higherthanthatofthecontrolgroup.Real-timereversetranscription#8211;polymerasechainreactionanalysisrevealedthatexposureofpreimplantationembryostoinsulinincreasedthemRNAexpressionofbothIgf2andH19inembryonicday(E)14fetuses.BisulfitegenomicsequencingdemonstratedthatthemethylationleveloftheH19#8211;Igf2imprintcontrolregionwas19.3%lowerininsulin-exposedE14fetusesthanincontrols.Thepresentstudyindicatesthatinsulinexposureduringthepreimplantationstagealterstheexpressionofimprintedgenesandaffectsfetaldevelopment.
