Noninvasive Assessment of Pulse-Wave Velocity and Flow-Mediated Vasodilation in Anesthetized G?ttingen Minipigs

Fewmethodsfornoninvasiveassessmentofarterialstiffnessandendothelialdysfunctioninporcinemodelsareavailable.TheaimofthisstudywastoevaluatemethodsforassessmentofarterialstiffnessandendothelialdysfunctioninanesthetizedG?ttingenminipigs.Pulse-wavevelocity(PWV)wasassessedinmaleG?ttingenminipigs(n=8;ageapproximately60wk)byusingapplanationtonometryofthecarotidandfemoralarteries.Inaddition,flow-mediatedvasodilation(FMD)wasassessedbyusingvascularultrasonographyofthebrachialarterytoevaluateendothelialdysfunction.Toevaluatethereproducibilityofthemethods,minipigswereanesthetizedbyintravenousinfusionofketamineandmidazolamandexaminedeveryotherdayforatotalof3trials.Neitherexaminationdaynorsystolic,diastolic,ormeanarterialbloodpressurestatisticallyinfluencedPWVorFMD.Themedianinterexaminationcoefficientofvariationwas17%forPWVand59%forFMD.MeasuredvaluesofPWVcorrespondedlargelytothoseinclinicallyhealthyhumans,butFMDvalueswerelowerthanexpectedforlean,younganimals.Althoughtheketamine#8211;midazolamanesthesiaweusedhasbeenassociatedwithminorhemodynamiceffectsinvivo,invitrostudiessuggestthatbothdrugsarevasodilatory.Thereforeanesthesiamighthaveinfluencedtheendothelialresponse,contributingtothemodestFMDresponseandtheconcurrenthighcoefficientsofvariationthatwenoted.WeconcludethatPWV#8212;butnotFMD#8212;showedacceptableinterexaminationvariationforitspotentialapplicationinporcinemodels.
