Wereportaserendipitousfindingofurinarycalculiinratsfedasemi-syntheticbasaldiet.ThisobservationwasmadeduringongoingstudiestoevaluatetheinhibitoryeffectofPSC833,apotentinhibitorofP-glycoprotein,ondevelopmentoftumorsinrodenttumormodelsystems.Alargenumberofspecific-pathogen-free(SPF)femaleSprague-DawleyandSPFmaleFischer344ratsbeingfedthedietwereeuthanizedwhenitbecameevidentclinicallythattheywereuremic.Atnecropsy,therenalpelvis,ureters,andurinarybladdercontainednumerouscalculi.ThepresenceofurinarycalculiwasdeterminedtoberelatedtothesourceofaFoodChemicalCodexgradeofcholinebitartrate.RatsbeingfedthesamebasaldietcontainingtheUnitedStatesPharmacopiagradeofcholinebitartratefailedtodevelopurinarycalculi.Interestingly,ratstreatedwiththeP-glycoproteininhibitorwereatsignificantlyreducedriskofdevelopingurinarycalculi.Thisfindinghighlightshowsomethingseeminglyinnocuousasaminordietaryconstituentcanhaveaprofoundimpactand,thereby,affectexperimentaloutcome.
