Woodisoftenusedasacontactbeddingmaterialforlaboratoryanimals.Ithasbeenestablishedthatwood,particularlysoftwood,hasthepotentialtoinducehepaticdrug-metabolizingenzymes.However,totheauthors'knowledge,changesinenzymeactivityafterremovalofanimalsfrombeddinghavenotbeencharacterized.Thepurposeofthestudyreportedherewastodeterminehowhepaticethoxyresorufin-O-deethylationandp-nitrophenolhydroxylationactivitiesalterovertimeafterremovalofratsfrompinebedding.Malerats,threetofourmonthsold,raisedincagescontainingpinebedding,weretransferredtowire-bottomedcages.Atvarioustimesthereafter(upto84days),groupsofratswereeuthanizedandtheliverwasprocessedtoobtainmicrosomes.ThemicrosomalproteinandtotalcytochromeP450(CYP)contentandenzymeactivitiesweredetermined.SignificantdifferencesintotalmicrosomalproteinortotalCYPvalueswerenotobservedoverthe84days,butadecreaseinethoxyresorufin-O-deethylationandp-nitrophenolhydroxylationactivitieswasdetected.Forp-nitrophenolhydroxylation,thedecreasewasexponential,withahalf-lifeofapproximatelyninedays,whereasforethoxyresorufin-O-deethylation,arapiddecreaseinactivityinthefirstweekwasfollowedbyareducedrateofdecreasethereafter.Enzymeactivitiesdidnotstabilizeforatleastsixweeks.Researchersusinglaboratoryanimalsshould,therefore,beawarethatittakesseveralweeksforenzymeactivitiestostabilizeonceanimalsareremovedfromthebedding.
