Ocularcolobomaissometimesaccompaniedbycorectopiainhumansandthereforeectopicpupilmayindicateocularcolobomainexperimentalanimals.TheRCSstrainofratshasalowincidenceofmicrophthalmia.Wefoundthatinferiorectopicpupilisassociatedexclusivelywithsmall-sizedeyesinthisstrain.TheobjectiveofthecurrentstudywastoevaluatewhetherinferiorectopicpupilisassociatedwithiridalcolobomaandothertypesofocularcolobomainRCSrats.BotheyesofRCSratswereexaminedclinically,andthosewithinferiorectopicpupilsunderwentmorphologicandmorphometricexaminations.Inaprenatalstudy,coronalserialsectionsofeyeballsfromfetusesatgestationalday16.5wereexaminedbyusinglightmicroscopy.EctopicpupilsinRCSratswerefoundexclusivelyinaninferiorposition,wheretheiriswasshortened.Fundicexaminationrevealedseverechorioretinalcolobomainallcasesofinferiorectopicpupil.Themorphologiccharacteristicscloselyresembledthoseofchorioretinalcolobomainhumans.Histopathologicexaminationofprimordiashowedincompleteclosureoftheopticfissurein4eyeballsofRCSfetuses.NeitherFsub1/subratsnorNsub2/sub(progenyofRCS×BNmatings)displayedanyocularanomalies,includingectopicpupils.TheRCSstrainisasuitablemodelforhumanocularcoloboma,andinferiorectopicpupilappearstobeastrongindicatorofocularcoloboma.
