MicelackingtheNHE2Na+/H+genedevelopgastritisoftheglandularmucosaasearlyasthetenthdayoflife,achievingmaximalintensityofinflammationfrom17to19daysafterbirthandmaximalatrophyatoneyear.Weassessedtheeffectsofthisprocessinsuchmiceto16monthsofage.ThestomachofNHE2nullmutantswasexaminedat10,17to20,24to35and49to70days,andat12to16months.TheNHE2wild-type(+/+)andNHE2heterozygous(+/-)micewerecomparedwiththeNHE2homozygousmutantmice(-/-).Thestomachofthemutantmiceatallageswascharacterizedbyasubstantiallyreducednumberofparietalcells.The10-day-oldmousestomachhadatransmuralinfiltrateofprimarilyneutrophils.Withincreasingage,neutrophilswerereplacedbylymphocytesandplasmacellsintheglandularmucosaofthemutantmice.Youngadult49-to70-day-oldmicehadsurfacecellhyperplasiaandexpansionofthereplicatingcellpopulation.Hyperplasiaofenterochromaffin-likecellsandantralgastrincellsaccompaniedprofoundfundicglandandsurfacecellhyperplasia,andbecameprogressivelymoreseverewithincreasingageoftheNHE2-/-mice.Neoplasmswerenotfoundinthemutantorcontrolmice.Thisgastritisdiffersfromthatofautoimmunegastritisinthatitistransmural,beginsininfancy,andisassociatedwithapredominantlyneutrophilicinfiltrateinitsearlystages.Someofthehistologicchangesintheadultmicecanbeexplainedonthebasisofprolongedachlorhydria.Thismousemaybeasuitablemodelforprolongedeffectsofachlorhydria.
