Mousepoxwasidentifiedinasinglemouse-holdingroominearly1999afteragroupof20CAF1/HsdmicewereinoculatedSCwithakilledmurinespindlecelltumorline,S1509A.Thecelllinehadbeenusedwithoutcomplicationsmultipletimesandwasdeterminedtobefreeofviralcontaminationonthebasisofresultsofmouseantibodyproductiontesting.Ofthe20miceinoculated,12micediedbypostinoculationday8.Severelymphoidandhepaticnecrosiswasobservedinselectmicesubjectedtohistologicexamination.Ballooningdegenerationofepithelialcellswithintracytoplasmiceosinophilicinclusionbodieswasobservedintheskinoverlyingtheinoculationsiteofthesinglemousefromwhichthistissuesitewasevaluated.Presenceofectromeliaviruswasconfirmedbyuseofimmunohistochemicalandpolymerasechainreactionanalyses,andtheviruswasisolatedafterserum,pooledfrom5oftheindexcases,wasinoculatedintoanimmune-naivemouse.InvestigationintothesourceofviruscontaminationincludedinoculatingmicewithaliquotsofvariousS1509Afreezedates;chemicallydefinedmediaandsupplements,includingfetalbovineserum;andtwolotsofpooledcommercialmousesera,afterheatinactivationat56°Cfor30minutesusedasamediumsupplement.Onelotofpooledcommercialmouseserumwasidentifiedasthesourceofectromeliavirus.ThislotofserumwasinadvertentlyusedtofeedS1509Acellsthatweresubsequentlyinoculatedintomice.Wedeterminedthatthecontaminatedserum,whichwaspurchasedinlate1998,originatedfromChina.TheserumwasimportedintotheUnitedStatesasabatchof43Linearly1995.Theserumwasblendedintoasinglelotandfiltered(0.#59157;2m)beforedistributiontomajorsuppliersthroughoutthecountry.Theserumwassoldorfurtherprocessedtoobtainavarietyofserum-derivedproducts.Becausemurineserumisgenerallysoldinsmallaliquots(10to50ml),wespeculatethatseveralthousandaliquotsmayhavebeenderivedfromthisbatchofserumand,ifinoculatedintomice,wouldlikelyresultinadditionalmousepoxoutbreaks.
