Successfulimmunereconstitutionwouldenhanceresistanceofbeige/scidmicetochronicinfectionwithMycobacteriumaviumsubspeciesparatuberculosis,butmaycausedamagetointestinaltissue.Therefore,weinvestigatedtheeffectofadoptivetransferofBALB/cmousesplenocytesonlesionseverityandintestinalphysiologyinbeige/scidmiceinfectedwithM.paratuberculosis.Micewereinoculatedintraperitoneally(i.p.)withM.paratuberculosis,andtwoweekslaterwereinoculatedi.p.withviablespleencellsfromimmune-competentBALB/cmice.Micewerenecropsied12weeksafterinfectionwhenengraftmentoflymphocytes,clinicaldisease,pathologiclesions,andintestinalelectrophysiologicparameterswereevaluated.Lymphocyteswererareincontrolbeige/scidmicenotinoculatedwithspleencells.Incontrast,highnumbersofCD4+,CD8+,andB220+lymphocytesweredetectedinthespleenofallbeige/scidmice(n=24)inoculatedwithspleencells,indicatingthatadoptivetransferresultedinsuccessfulengraftmentofdonorlymphocytes(immunereconstitution).ImmunereconstitutionofM.paratuberculosis-infectedbeige/scidmicesignificantlyreducedtheseverityofclinicaldiseaseandpathologiclesions,andnumbersofbacteriaintheliver.However,intestinalelectrophysiologicparametersstudiedinvitroindicatedthatintestinaltissuesfromreconstitutedbeige/scidmicehadreducedshort-circuitcurrentresponses(duetoreducedionsecretion)followingelectrical,glucose,andforskolinstimulation.Theseabnormalresponsessuggestedthatneuralorepithelialcellsintheintestineweredamaged.Weconcludethatsuccessfulimmunereconstitutionofbeige/scidmiceenhancetheirresistancetoM.paratuberculosisinfection,butmaycausepathophysiologicchangesassociatedwithintestinalinflammation.
