BackgroundandPurpose:Validationofamethodforobtainingbloodsamplesthatdoesnotchangecortisolorprolactinconcentrationsyetallowsserialbloodsamplestobecollectedfromanimalsunderanesthesia,withoutpriorhandling,fromfreelyinteractingsocialgroupsofsmallmammals.brxmlns="http://pub2web.metastore.ingenta.coms/"/brMethods:Resultsfromfiveexperimentsarereported.Maledwarfhamsters(Phodopusspp.)werehousedinmodifiedhomecagesundercontinuousflowofcompressedairthatcouldbeswitchedtoisofluraneinOsubxmlns="http://pub2web.metastore.ingenta.coms/"2/subvehiclewithoutapproachingthecages.brxmlns="http://pub2web.metastore.ingenta.coms/"/brResults:Dwarfhamstersrespondtomanualrestraintwithbehavioraldistressandincreaseintheconcentrationofthedominantglucocorticoid,cortisol,anddecreaseinprolactinconcentration.Botheffectsareevidentwithinoneminute.Incontrast,whenthisnewmethodwasused,neithercortisolnorprolactinchangedinresponsetorepeatedsamplecollection(upto8successivesamplesat2hourintervals),prolongedisofluraneexposure,orsubstantialbloodvolumereduction(30%).Prolactinconcentrationwassuppressedandcortisolconcentrationwasincreasedinresponsetostimulifromotherhamsterstestedwithoutanesthesia.Suppressionofprolactinconcentrationwasgradedinresponsetothedegreeofstressandequaledthepharmacologicreductioncausedbybromocryptinemesylate(50#59157;gofCB154x3days).brxmlns="http://pub2web.metastore.ingenta.coms/"/brConclusions:Thetechniqueissuperiortoalternativesforstudiesofbehavioralendocrinologyoffreelyinteractingsmallmammals.
