Trendsingeneticsaretransforminginordertoidentifydifferentialcoexpressionsofcorrelatedgeneexpressionratherthanthesignificantindividualgene.Moreover,itisknownthatacombinedbiomarkerpatternimprovesthediscriminationofaspecificcancer.Theidentificationofthecombinedbiomarkerisalsonecessaryfortheearlydetectionofinvasiveoralsquamouscellcarcinoma(OSCC).ToidentifythecombinedbiomarkerthatcouldimprovethediscriminationofOSCC,weexploredanappropriatenumberofgenesinacombinedgenesetinordertoattainthehighestlevelofaccuracy.Afterdetectingasignificantgeneset,includingthepre-definednumberofgenes,acombinedexpressionwasidentifiedusingtheweightsofgenesinageneset.WeusedthePrincipalComponentAnalysis(PCA)fortheweightcalculation.Inthisprocess,weusedthreepublicmicroarraydatasets.Onedatasetwasusedforidentifyingthecombinedbiomarker,andtheothertwodatasetswereusedforvalidation.Thediscriminationaccuracywasmeasuredbytheout-of-bag(OOB)error.Therewasnorelationbetweenthesignificanceandthediscriminationaccuracyineachindividualgene.Theidentifiedgenesetincludedbothsignificantandinsignificantgenes.OneofthemostsignificantgenesetsintheclassificationofnormalandOSCCincludedemMMP1/em,emSOCS3/emandemACOX1/em.Furthermore,inthecaseoforaldysplasiaandOSCCdiscrimination,twocombinedbiomarkerswereidentified.Thecombinedexpressionrevealedgoodperformanceinthevalidationdatasets.Thecombinedgenomicexpressionachievedbetterperformanceinthediscriminationofdifferentconditionsthanasinglesignificantgene.Therefore,itcouldbeexpectedthataccuratediagnosisforcancercouldbepossiblewithacombinedbiomarker./p/div
