Histonemodification(HM)patternsarewidelyappliedtoidentifytranscriptionfactorbindingregions(TFBRs).However,howfrequentlytheTFBRsoverlapwithgenomicregionsenrichedwithcertaintypesofHMsandwhichHMmarkerismoreeffectivetopinpointtheTFBRshavenotbeensystematicallyinvestigated.Toaddresstheseproblems,westudied149transcriptionfactor(TF)ChIP-seqdatasetsand33HMChIP-seqdatasetsinthreecelllines.Wefoundthatonaverageabout90%oftheTFBRsoverlapwiththeH3K4me2-enrichedregions.Moreover,theH3K4me2-enrichedregionswithstrongersignalsofH3K4me2enrichmentmorelikelyoverlapwiththeTFBRsthanthosewithweakersignals.Inaddition,weshowedthattheH3K4me2-enrichedregionstogetherwiththeH3K27ac-enrichedregionscangreatlyreducefalsepositivepredictionsoftheTFBRs.OurstudyshedslightonthecomprehensivediscoveryoftheTFBRsusingtheHeK4me-enrichedregions,especiallywhennogoodantibodytoaTFexists./p/div
