Non-alcoholicfattyliverdisease(NAFLD)isthemostpredominantliverdiseaseworldwideandhepaticmanifestationofthemetabolicsyndrome.Itshistologyspectrumrangesfromsteatosis,tosteatohepatitis(NASH)thatcanfurtherprogresstocirrhosisandhepatocellularcarcinoma(HCC).TheincreasingincidenceofNAFLDhascontributedtorisingnumbersofHCCoccurrences.NAFLDprogressionisgovernedbygeneticsusceptibility,environmentalfactors,lifestyleandfeaturesofthemetabolicsyndrome,manyofwhichoverlapwithHCC.GeneexpressionprofilingandgenomewideassociationstudieshaveidentifiednoveldiseasepathwaysandpolymorphismsingenesthatmaybepotentialbiomarkersofNAFLDprogression.However,themultifactorialnatureofNAFLDandthelimitednumberofsufficientlypoweredstudiesareamongthecurrentlimitationsforvalidatedbiomarkersofclinicalutility.Furtherstudiesincorporatingthelinksbetweencircadianregulationandhepaticmetabolismmightrepresentanadditionaldirectioninthesearchforpredictivebiomarkersofliverdiseaseprogressionandtreatmentoutcomes./p/div
