Modified Yersinia pestis strains with a favourable safety profile have been shown to confer long-term protection against pneumonic plague. Ashok Chopra and colleagues at the University of Texas genetically modified a strain of Y. pestis—the pathogen causing plague-deleting the genes that allow the bacterium to proliferate in a host and cause disease. Using rat and mouse models, the team found their mutated strains caused no pathogenic symptoms yet generated total immunity when challenged with the live bacteria at 120 days post—immunization, with protected animals expressing significantly higher levels of immune system mediators. The findings call for investigation into the suitability of this vaccine in humans due to the lack of an FDA-approved vaccine for plague, with current vaccines causing severe side effects and death if used by certain individuals.
