Bcells,asanimportantpartofthehumoralimmuneresponse,aregeneratedintheBM,migratetosecondarylymphoidorgans,anduponactivation,differentiateintoantibody‐producingmemoryBcellsorplasmacells.Despitethepivotalrolesthattheyplayindifferentdiseases,acomprehensivecharacterizationinhealthyrhesusmacaques,whichserveasvaluablemodelsforavarietyofhumandiseases,isstillmissing.Withtheuseofmultiparameterflowcytometry,weanalyzedBcellsinBMcollectedfromtwolocations,i.e.,theiliaccrest(BMca)andthefemur(BMfem),PB,aswellassecondarylymphoidorgansofhealthyrhesusmacaques.WeassessedthefrequenciesofimmatureandmatureBcells,aswellasCD19+CD20–CD38+/++CD138+/++plasmablasts/plasmacells.Furthermore,wefoundsite‐specificdifferencesintheexpressionofmarkersforBcellactivationandproliferation,chemokinereceptorsandIgs,aswellasthedistributionofmemoryBcellsubpopulations.AssecondarylymphoidorgansharborthehighestfrequenciesofnaiveBcells,expressionofCD80,CD95,andKi67waslowercomparedwithBcellsintheperipheryandBM,whereasexpressionofIgD,CXCR4(CD184),andCCR7(CD197)washigher.Interestingly,BMcadifferedfromBMfemregardingfrequenciesofBcells,theirexpressionofCD80andCXCR4,Tcells,andplasmacells.Insummary,thesedataidentifybaselinevaluesfortheabove‐mentionedparametersandprovidethefoundationforfuturestudiesonBandplasmacellsindifferentdiseases...
