Wehaveexaminedthecholinergicsympatheticinnervationofsweatglandsinfootpadsofadultanddevelopingrats.Acetylcholinesterasestainingrevealsaplexusofheavilystainedfibersinthesweatglandsofadultrats.Reactionproductappearsamongandaroundbundlesofaxonsthatlieataconsiderabledistancefromthecellsofthesecretorytubule.Eachbundlecontains8a€“12axonsthatpossessnumerousvaricositiesandcontainsmallclearandlargedensecorevesicles.Theglandsofthehindpawsandtheirinnervationdevelopduringthefirstthreeweeksafterbirth.Catecholamine-containingaxonswereassociatedwiththeformingglands.At7and10days,intenselyfluorescentfiberssurroundedthetubules,andalloftheaxonprofilesassociatedwiththeglandscontainedsmallgranularvesicles(SGV)afterpermanganatefixationtorevealvesicularstoresofnorepinephrine.At14daysthesweatglandplexuswaslessintenselyfluorescentthanatearlieragesandrelativelyfewSGVwerepresent.By21days,noendogenouscatecholaminefluorescenceandnoSGVweredetectable.However,followingexposuretoexogenouscatecholamine,fluorescentfiberswerepresentinthesweatglandsofmatureratsandtheycorrespondedinpositionanddensitytotheplexuslocalizedwithacetylcholinesterasestaining.Catecholamineuptakewasblockedbyincubationinthecoldandbydesmethylimipramineandwasnotobservedincholinergicparasympatheticfibersintheirisorsalivaryglands.Afterintraperitonealadministrationof5-hydroxydopamineandpermanganatefixation,alltheaxonsinthesweatglandscontainedafewSGV.Thus,thedevelopingsweatglandsappeartobeinnervatedbynoradrenergicaxonsthatlosetheirstoresofendogenouscatecholaminesbutnottheircapacityforuptakeandstorageastheyelaborteanaxonalplexusinthematuringglands.Theseobservationssupportthehypothesisthatcholinergicsympatheticneuronsappeartoundergoatransitionfromnoradrenergictocholinergicfunctionduringdevelopmentinvivosimilartothatpreviouslydescribedincellculture.
