Depression of reproduction or embryonic death can be observed in intersubspecific hybrids after 2nd generation of mating in mice. This 'hybrid breakdown' may be due to genetic incompatibilities caused by nucleotide polymorphisms found in genes of both subspecies. Such genetic incompatibilities are thought to affect those reproduction or embryogenesis in hybrids. Here we carried out sib (sister - brother)-mating experiments using offspring from two different subspecies, which were C57BL/6J (M. m. domesticus) and MSM/Ms (M m. molossinus). The mating was continued to the 5th generation. We found that their litter size continuously declined as the generation number increased in several different families. There were significant differences in their median litter size per delivery between the 1st and the 5th generation. We also found that number of ovulated eggs decreased significantly in the 5th generation compared to the 1st generation individuals. Ovaries of poor morphology were specifically found in 5th generation. It was previously reported that hybrid breakdown / sterility was caused by abnormalities in spermatogenesis. Here we suggest that reduction of reproductive ability in females is also a factor of hybrid breakdown.
