Successful microinsemination of rabbit oocytes with round spermatids

Although the first report for rabbit offspring by round spermatid injection (ROSI) was published in 1994 (Sofikitis et al.), the production of rabbit ROSI offspring has been considered difficult due to high chromosomal abnormality (Ogonuki et al., 2004). The present study was designed to investigate the effect of oocyte activation regimens on developmental ability of rabbit ROSI oocytes. Denuded kbs: JW rabbit oocytes were treated with 5-10 μM ionomycin for 5 min before ROSI (Group-1), after ROSI (Group-2), before and after ROSI (Group-3), and before and after ROSI followed by 1-h incubation with 5 μg/ml cycloheximide + 2 mM 6-dimethylaminopurine (Group-4). Intracytoplasmic sperm injection (ICSI) was performed using non-treated oocytes (Group-5). The activation regimens influenced the proportions of ROSI oocytes cleaving after 22 to 24 h of culture (39, 29, 51, 68 and 60% in Group-1, -2, -3, -4 and-5, respectively). Oviductal transfer of the cleaved embryos resulted in implantation signs in Group-3 (2%) and Group-4 (9%), and full-term development only in Group-4 (7%). The ICSI resulted in 60% cleavage, 4% implantation and 8% full-term development (Group-5). Thus, rabbit ROSI oocytes were capable of developing into full-term when the oocytes were activated with a combined treatment of ionomycin and cycloheximide + 6-dimethylaminopurine.
