In our institute, we started mouse embryo cryopreservation in 2004, microbiological cleaning and recovery of cryopreserved embryos in 2005 as a duty. We report the requests and the results of these duties from 2004 to now. Mouse embryo cryopreservation was carried out according to simple vitrification procedure. To examine a viability of cryopreserved embryos, some cryopreserved embryos were recovered and developed. When over 200 embryos were cryopereserved and over 30% of cryopreserved embryos were developed, we finished embryo cryopreservation. To clean up the microbes, 2-cell stage embryo transplanted to the recipient mice. After the transplantation the recipient mice were housed in isolator. After weaning the microbes in the recipient mice were monitored. And if the recipient mice were free from the microbes, we finished microbiological cleaning. Recovery of cryopreservated embryos was carried out as usual. Because embryo cryopreservation could be finished in 24 lines of mice, we have no problem in our cryopreservation method. But we needed to examine the viability of cryopreserved embryos again in the 11 lines therefore we have some problems in our examination techniques. And we have no problem in microbiological cleaning and recovery of cryopreserved embryos in our institute. From these results, we can state that our duties of the manipulating mouse embryo are smoothly carried out.
