摘要

We develop a high-throughput screening method to distinguish between wild type (WT) and mutant Arabidopsis plants based on the deviation in their cell wall polysaccharide structure. For the set-up of this screening method we have chosen a mutant plant which is deficient in xyloga-lacturonan (xgd1) and a WT plant (eco-type Co10). We degraded the cell wall material of leaves from the WT and mutant plant with different mixtures of pure and well denned enzymes: pectinases (including polygalacturonase, pectinmethyleste-rase, rhamnogalacturonan hydrolase and xylogalacturonan hydrolase), hemicellu-lases (including mannanase, galactanase, arabinanase, xylanase, glucanaseV) and cellulose degrading enzymes (glucanasel, cellobiohydrolase).
