Drugmakers like to brag about the latest high-tech tools―computerized gene sequencers that unravel DNA, automated machines that screen thousands of compounds in hours―and how they will lead to a boatload of breakthroughs. In reality the difference between spectacular success and abject failure can come down to a little luck and a few dedicated inventors toiling behind the scenes. These medical Merlins have the rare combination of genius, drive and faith needed to surmount dismal odds against a new drug's making it to market. Some are brilliant biologists who uncloak the role of crucial disease-causing genes. Others are crafty chemists who concoct compounds to safely block those bad genes. And still others are tenacious proselytizers who keep a project alive when no one else believes. Here's a look at five of them.
