Gradient diffusion and protein solubility: use of dynamic light scattering to localize crystallization conditions

The gradient diffusivity, D, of proteins is measured with conventional dynamic light scattering instruments. In the low volume fraction limit, D is a linear function of volume fraction and can be written: D/D_0 = 1 + D_2 φ, where D_0 is the limiting low volume fraction diffusivity and D_2 characterizes the pair contributions of hydrodynamic drag and osmotic compressibility on diffusivity. As the strength of particle attraction increases, D_2 becomes increasingly negative. Here we demonstrate that the sensitivity of D_2 to attractions can be used as a method to guide in the location of the crystallization slot and suggest a titration method that can be used to rapidly identify solution conditions placing the protein within this slot.
