ASSESSMENT OF EMBRYOGENIC CALLI DERIVED FROM TEMPERATURE SENSITIVE GENIC MALE STERILITY BASED RICE HYBRIDS FOR SALT TOLERANCE

Five temperature sensitive genie male sterile (TGMS) lines viz., GD 98028, GD 98029, GD 98021, GD 98168 and GD 98179, eight salt tolerant testers viz., CSR 10, CSR 23, CSSRI 13, CSSRI 60, Nona Bokra, Vytilla 3, TRY 2 and CO 43 and 40 hybrids obtained by crossing them in Line x Tester design were screened for their salt tolerance under in vitro conditions. Earlier callus induction, higher percentage of callus induction and maximum fresh weight of callus was obtained when rice embryos were inoculated and cultured in MS media supplemented with 2.4-D 2 mg l~(-1), kinetin 0.25 g l~(-1) and casein hydrolysate 1 g l~(-1). The media were utilized in screening of parents and hybrids for salt tolerance in five varied concentrations of NaCl (0.0,0.4,0.8,1.2 & 1.6 %). The genotypes were ranked based on the criteria callus morphology score and fresh weight of callus recorded after three weeks of screening. The hybrid GD 98029/CSSRI 13 ranked first followed by the hybrids GD 98028/CO 43, GD 98028/CSR 23, GD 98029/CSR 10, GD 98029/Nona Bokra and the parent Nona Bokra in MS media supplemented with 2,4-D 2 mg l~(-1), kinetin 0.25 g l~(-1) and casein hydrolysate 1 g l~(-1) along with NaCl at 1.6 per cent concentration exhibiting their potential for salt tolerance.
