Hypersensitivity to low-dose of ionising radiation (HRS) is a well-documented phenomenon that illustrates well a biological non-linearly dose dependent effect where the molecular and cellular events observed in a low-dose range (generally from 1 mGy to 0.8 Gy) are similar to those observed in 5-50 times higher doses. Up to now, the HRS phenomenon remained unexplained. Since 2003, we accumulated a large number of individual radiosensitivity cases whether well characterized genetically or revealed after postradiotherapy tissue reactions: after a dose of 2 Gy (one standard radiotherapy session), we pointed out that the ATM protein shuttles from cytoplasm to nucleus to activate the DNA double-strand breaks repair. The more efficient the transit, the higher the radioresistance. Here, we provided evidence that the transit of ATM can explain the HRS phenomenon, whose consideration may change the estimation of the risk linked to low-dose radiation.
