摘要

Large parts of mammalian genomes are transcriptionally inactive and enriched with various classes of interspersed and tandem repeats. Here, we showed that the tumor-suppressor protein p53 cooperates with DNA methylation to maintain transcriptional silencing of these parts of the mouse genome. Furthermore, failure of p53 and DNA methylation to maintain epigenetic silencing of repeats results in massive accumulation of new RNA species, which trigger a suicidal interferon (IFN) response that eliminates cells with unrepressed repeats.
