Cancer cells experience compressive stress as they undergo uncontrolled proliferation in the body's confined spaces. To determine how these stresses affect tumor cells, Janet Tse et al. (pp. 911-916) subjected mammary epithelial cells to mechanical compression by pressing them against a membrane surface with a weighted piston. The authors found that compressive stress enhanced the migration of mammary carcinoma cells by stimulating the formation of "leader cells" at the leading edge of cell sheets. Leader cells extend protrusions in the direction of migration while maintaining connections with trailing follower cells. Compression also induced cells to secrete a protein called fibronectin that improves migration and leader cell formation by increasing the adhesion between cells and the surface.
