KEY MOLECULAR EVENTS IN CARCINOGENESIS

Cancers Develop through a multistage process which can encompass an appreciable part of lifespan of the species (Weinstein et al 1995, Hanahan & Weinberg 2000). Remarkable progress has been made in our understanding of the cellular, biochemical and molecular genetic events that occur during the multistage process. Uncontrolled cell proliferation is one of the main hallmarks of cancer, and tumor cells acquire damage to genes that are directly involved in regulating the cell cycle (Sherr 1996, Weinstein 2000). The step-wise process of carcinogenesis reflects the progressive acquisition of mutations producing oncogenes with dominant gain of function, and of mutations in tumor-suppressor genes with recessive loss of function (Vogelstin & Kinzler, 1998). Altered gene products lead to activation in signal transduction resulting in modified gene expression. Epigenetic abnormalities in the expression of oncogenes and tumor-suppressor genes also play an important role in carcinogenesis. Regardless of the genetic damage or type of cancer, the common feature is a disrupted cell cycle.
