Cryopreservation of Plant Germplasm: A Review (Scientific Review)

Cryopreservation has been adopted to store base collections of plant material in liquid N_2 in order to ensure safe, and cost-effective long-term conservation of plant genetic resources. The Cryopreservation of plant material has been accomplished bymeans of several techniques, including two-step freezing, vitrification, encapsulation-dehydration, and encapsulation-vitrification. Cryopreservation must avoid the formation of ice crystals inside cells during immersion in liquid N_2 which can be achieved by lowering cell water content prior to rapid cooling. Treatments that result in low cell water content are key factors for successful Cryopreservation. Several pretreatments, including cold acclimation, exposure to abscistc acid (ABA), immersion in concentrated sugar solution or osmoticums, and air dehydration have been considered to enhance the cryopreser vability of plant material. Plant Cryopreservation protocols must include the establishment of a reliable recovery and regrowth procedures afterCryopreservation.
